- Ciliocytophthoria in cervical cytologyPublication . Mendonça, Paula; Albuquerque, M.; Antunes, L.; Gonçalves, A.; Pinheiro, C.; Marcos, F.The Ciliocytophthoria occurs in the glandular epithelium/cylindrical ciliated cells, which are found in the respiratory and gynaecological systems. Although this phenomenon is considered rare in both systems, it is more unusual in the gynaecological system. Of the 5781 cases analysed at the centre in 2006, only one case was diagnosed as Ciliocytophthoria. In 1975, Kobold-Wolterbeek and Beyer-Boon observed the cervical smears of 18 000 women during a period of a year and a half, during which time a total of 32 cases of Ciliocytophthoria were found. This phenomenon consists of the separation of the cylindrical cells resulting in the formation of a non-nucleus portion – terminal plate with cilia – and a nucleus portion with granular cytoplasm containing eosinophilia inclusions. The relationship between Ciliocytophthoria and any disease or tumour in the female genital tract is unknown.
- Optimization of immunocytochemistry in cytology: comparison of two protocols for fixation and preservation on cytospin and smear preparationsPublication . Pinheiro, C.; Roque, Rúben; Adriano, A.; Mendes, P.; Praça, M.; Reis, I.; Pereira, T.; Srebotnik Kirbis, I.; André, S.Objective: A new protocol for fixation and slide preservation was evaluated in order to improve the quality of immunocytochemical reactions on cytology slides. Methods: The quality of immunoreactions was evaluated retrospectively on 186 cytology slides (130 direct smears, 56 cytospins) prepared from different cytology samples. Ninety-three of the slides were air dried, stored at -20 °C and fixed in acetone for 10 minutes (Protocol 1), whereas the other 93 were immediately fixed in methanol at -20 °C for at least 30 minutes, subsequently protected with polyethylene glycol (PEG) and stored at room temperature (Protocol 2). Immunocytochemical staining, with eight primary antibodies, was performed on a Ventana BenchMark Ultra instrument using an UltraView Universal DAB Detection Kit. The following parameters were evaluated for each immunoreaction: morphology preservation, intensity of specific staining, background and counterstain. The slides were blinded and independently scored by four observers with marks from 0 to 20. Results: The quality of immunoreactions was better on methanol-fixed slides protected with PEG than on air-dried slides stored in the freezer: X¯ = 14.44 ± 3.58 versus X¯ = 11.02 ± 3.86, respectively (P < 0.001). Conclusion: Immediate fixation of cytology slides in cold methanol with subsequent application of PEG is an easy and straightforward procedure that improves the quality of immunocytochemical reactions and allows the storage of the slides at room temperature.

F.; Medrado, L.Os SIGs são Grupos Especiais de Interesse (SIG - Special Interest Group) que buscam promover a integração e troca de experiências entre Unidades e Núcleos de Telessaúde membros da Rede Universitária de Telemedicina (RUTE), um programa da Rede Nacional de Ensino e Pesquisa (RNP/MCTI). A meta do grupo é a interação entre profissionais de citotecnologia de países de língua portuguesa para promover capacitações, discutir projetos conjuntos e a publicação de materiais de ensino na área. O objetivo deste documento foi definir as competências a adquirir pelos estudantes na citecnologia (formação básica e/ou avançada) de modo a que as entidades formadoras tenham uma orientação para a elaboração e atualização de programas formativos dirigido à área de conhecimento da citotecnologia.
